Artesunate + Amodiaquine Combination Antimalarial

 


COMPOSITION: Artesunate 100mg + Amodiaquine 153mg base

 

FORMULATION: Tablets

 

DESCRIPTION:

Each package includes 6 artesunate tablets and 12 amodiaquine tablets.

 

INDICATIONS:

1.        Treatment of all forms of malaria i.e. falciparum, Vivax and malariae, etc.

2.        Treatment of malaria caused by resistant strain of plasmodium falciparum against other anti-malarial drugs (chloroquine etc.)

3.        Use in malaria control programmes.

 

Pharmacology:

Combination therapy on all forms of malaria.

After oral administration, amodiaquine hydrochloride is rapidly absorbed 4-aminoquinoline derivatives appear to bind to nucleproteins and inhibit DNA and RNA polymerase, and undergoes rapid and extensive metabolism to desethylamodiaquine which concentrates in blood cells.

Artesunate has an excellent anti-malarial activity on the asexual forms of malaria parasites with high, quick efficacy to bring about rapid control of acute malaria attack. After oral administration, artesunate is completely and rapidly absorption, widely distribution and thorough excretion. It has no cross resistance to Chloroquine.

The combination of Artesunate and Amodiaquine is sage and well tolerated. It results in a faster relief of symptoms than monotherapy and in a quicker parasites clearance compared to amodiaquine alone. And the combination is well tolerated and efficacious. The cure rate exceeds 90% in 14 days.

 

Dosage and administration:

By oral route for patients of all age groups

Adult (£¾14 years) patients should take 2 tablets of artesunate and 4 tablets of amodiaquine once daily for 3 days.

The recommended dosage for other age groups are as following, but be sure to ask advice from your physician or pharmacist before using it.

 

PRECAUTIONS: 

Pregnancy

Embryo-toxic effects of artesunate were observed in some animal studies. This drug should be used carefully in pregnant women during the first trimester.

 

 Lactation

No bile excretion of artesunate was reported in lactation patients. But report on amodiaquine is lack of. Use this drug carefully for lactation patients.

 

Interactions

There have been no reports of negative drug interactions to date. The activity of other anti-malaria drugs may in fact be potent.

 

Side effects

Few cases of side effects at the therapeutic dose were observed, mainly caused by amodiaquine. Abdominal cramps and mild diarrhea were the mostly familiar. And reticulocytopenia is another familiar side effect.

 

Over dosage

Do not exceed the recommended dosage without further medical advice.

In case of overdose, emergency symptomatic treatment in a specialist facility is required.

 

PRESENTATION:

Pack of 6 tablets of artesunate and 12 tablet of Amodiaquine.

 

STORAGE:

The tablets should preferably be kept dry and stored at room temperature. The drugs should always be kept in their original package.

Three years.
